چکیده
In this study, a potential novel cellulolytic bacterium Arthrobacter sp. HPG166 was isolated from the hindgut of root-feeding larvae Holotrichia parallela. Optimization of fermentation factors for endoglucanase production by Arthrobacter sp. HPG166 was carried out via response surface methodology. Sodium carboxymethylcellulose 1.19% (w/v) and beef extract 0.35% (w/v) were the ideal combination of carbon and nitrogen sources for enzyme production; the optimum temperature and pH for cellulase production were 34°C and pH 8.0 respectively. Under the optimized fermentation conditions, the maximum endoglucanase activity of 1.411 U mL -1 was obtained. The crude endoglucanase was thermotolerant as it retained 50.31% of its activity after incubation at 70°C for an hour. Metal profile of the enzyme indicated that Mg 2+ and Na + were strong stimulators while Mn 2+ and Co + drastically inhibited its activity. Due to its particular characteristics, this enzyme could have potential for industrial applications.

متن کاملA novel low molecular weight extracellular protease from a moderately halophilic bacterium Salinivibrio sp. strain MS-7: production and biochemical properties
Kinetics of bacterial growth and protease production were monitored on a novel isolated moderately halophilic bacterium, Salinivibrio sp. strain MS-7, and maximum growth and protease activity was achieved after 48 hours at 30°C and 180 rpm. To determine the effect of various carbon sources on protease production, glucose, lactose, sucrose and maltose were investigated and maximum production of...
متن کاملProduction and characterization of biochemical properties of L-Asparaginase by indigenous yeast isolated from soil of Iran
Introduction: The use of L-asparaginase as a chemotherapeutic agent is an effective strategy for the treatment of leukemic lymphoblasts. Since the use of bacterial L-asparaginase causes side effects due to hypersensitivity reactions, search for new enzyme sources is one of the active research areas. In this study, the characterization of produced L-asparaginase by indigenous yeast strains was i...

Background: Alginate is a linear polysaccharide consisting of guluronate (polyG) and mannuronate (polyM) subunits. Methods: In the initial screening of alginate-degrading bacteria from soil, 10 isolates were able to grow on minimal medium containing alginate. The optimization of cell growth and alginate lyase (algL) production was carried out by the addition of 0.8% alginate and 0.2-0.3 M NaCl ...
